  1. Levamisole

Levamisole, marketed as the hydrochloride salt under the trade name Ergamisol, is an anthelminthic and immunomodulator belonging to a class of synthetic imidazothiazole derivatives. It was discovered at Janssen Pharmaceutica in Belgium, in 1966, where it was prepared initially in the form of its racemate. The two stereoisomers of tetramisole were subsequently synthesized, and the levorotatory isomer was given the name levamisole. Levamisole has been used in humans to treat parasitic worm infections, and has been studied in combination with other forms of chemotherapy for colon cancer, melanoma, and head and neck cancer. In some of the leukemic cell line studies, both levamisole and tetramisole showed similar effect. The drug was withdrawn from the US and Canadian markets in 2000 and 2003, respectively, due to the risk of serious side effects and the availability of more effective replacement medications. The key toxic effect of the drug is agranulocytosis, a severe depletion of white blood cells that leaves patients vulnerable to infection. Currently, levamisole remains in veterinary use as a dewormer for livestock. The medication has also been increasingly used as an adulterant in cocaine sold in the US and Canada, resulting in serious side effects.

  1. Levamisole

An antihelminthic drug that has been tried experimentally in rheumatic disorders where it apparently restores the immune response by increasing macrophage chemotaxis and T-lymphocyte function. Paradoxically, this immune enhancement appears to be beneficial in rheumatoid arthritis where dermatitis, leukopenia, and thrombocytopenia, and nausea and vomiting have been reported as side effects. (From Smith and Reynard, Textbook of Pharmacology, 1991, p435-6)
